E. coli transformation
- thaw competent cells on ice
- mix 100 µl competent cells with your ligation reaction in 1.5 ml tube (or use down to 1/10 of your ligation)
- incubate on ice for 30 min
- heat shock at 42 °C for 1 min 45 sec
- add 400 µl 2 x TY
- incubate on shaker for 30 - 60 min at 37 °C
- plate 50 - 200 µl on selection plate
- incubate at 37 °C
