
Preparation of
Yeasts for FACS-Sytox
- Spin down 1.5 ml of cells from O/N culture
- Resuspend in 1 ml 70% or 100% EtOH and
rotate O/N
- Spin out of EtOH
- Resuspend in 1 ml H2O
- Spin and resuspend in 0.5 ml 50 mM Tris
pH 8.0
- Add 10 µ l RNase A (10 mg/ml), incubate
2 - 4 hrs at 37°C
- Spin and resuspend in 0.5 ml 50 mM Tris
pH 7.5 containing 2 mg/ml proteinase K
- Incubate 30 - 60 min at 50°C
- Spin out of protease solution
- Resuspend in 0.5 ml FACS buffer
- Transfer 100 µ l (or more if low
# of cells) to another tube, add 1 ml of 1X Sytox (stock is 5000x)
in 50 mM Tris pH 7.5
- Sonicate at setting 3 for 4 sec then analyze
Buffers:
FACS-buffer:
200 mM Tris/HCl pH 7.5; 200 mM NaCl; 78 mM MgCl2.
5000x
Sytox Solution:
5 mM Sytox Green in DMSO (Molecular
Probes S-7020)
